Main Lab
The CMI Core Main Lab occupies 650 sq. ft at the URI Providence Feisntein Campus. The core is responsible for providing 5 main services to TRIAD researchers.
Flow Cytometry – This is accomplished using a BD LSRII flow cytometer with a 3 laser package allowing for 9 color analysis. The BD LSRII also supports 96 well and 384 well assay formats, as well as individual tubes utilizing the BD High Throughput System The current optics package on the BD LSR II is made up of 3 lasers:
- 488 nm Blue Laser supporting FSC, SSC, FITC, PE (Pe Cy-7), PerCP (PerCP Cy-5.5, PerCP Cy7)
- 633 nm Red Laser supporting APC, APC Cy-7 (APC h-7)
- 405 nm Violet Laser supporting Pacific Blue, AmCyan
- Other custom fluorphores are supported such as alexa fluor stains, please inquire if you have any questions as to fluorophore selection.
ELISpot Assay Support – This is accomplished using a CTL Ltd. S5 UV Analyzer which is comprised of a precision microscope and digital camera package that can scan and capture images of individual ELISpot wells and Immunospot 5.0 which is a software package capable of automatically reading and analyzing spot size, density and intensity.
Cryogenic Storage – The core provides the ability to stably freeze cell and tissue culture samples at a controlled rate optimized for cell recovery using a Thermo Cryomed 7450 Controlled Rate Freezer, and maintains long term ultra low temp storage using a Thermo Cryoplus 7400 LN2 Dewar which has the capability iof storing up to 6800 2 ml vials.
Liposome Formulation – The preparation of liposome encased peptides, proteins or DNA constructs is becoming available in conjunction with Dr. Geoff Bothun of URI’s Chemical Engineering Department in Kingston. Sample preparation in Providence is performed using a Buchi R114 Rotovap, a Misonix 2000 sonicator and a Labconco Freezone 1 L Lyopholizer. Characterization is currently being perfoemd in the Bothun Lab in Kingston.
Transgenic Mouse Colony Maintenance – The core maintains 3 human transgenic mouse colonies currently at Taconic Farms in upstate New York. We are currently maintaining A2DR1, DR3 and DR4 mouse lines.

BSLII Animal Facility
Located at Peckham Farm on the University of Rhode Island’s Kingston campus, the BSLII animal facility is a 400 sq, ft lab equipped with state of the art Optimice Animal Cages, 2 Biosafety cabinets, CO2 Incubators, refrigeration, ultra low temp freezer and autoclave.
